Does the added PNG affect rendering?

Good afternoon!
Can you please tell me how true this assumption is?
1. There is a video, it lasts 1.5 hours, rendering took 2 hours.
2. There is a similar video, but it lasts 2.5 hours. A PNG strip has been added at the bottom of the video to close the taskbar. Rendering will last up to 10 hours.
Can the added PNG-scale affect the rendering so much or does Premier pro care about such trifles?
