Browser 2D / 3D games can be made on the Godot engine (perhaps in Unity too), there is an export for different platforms, including the browser (the output is html with js resources, sprites, sounds). You just need to be careful with heavy graphics, compress so that the browser does not slow down.
By the way, there are examples of Tower Defense on the github both in pure js and under Godot.
With the help of any broadcast in WebAssembly, for example, Unreal Engine added support for working in browsers three years ago.
