Video card not working in the second PCI-E x16 slot?

Hello, I have two video cards, sometimes I have to use the second one, but this time something went wrong. I inserted a second card (not two at the same time) into the upper slot and the PC began to beep 5 short-pause-1 short (or long, I did not understand). Prior to this, the card worked fine in the first slot, and it works, but if you insert it into the second PCI-E x16 slot. What does the first slot work 100% because. The first card there works fine. What could be the problem?
