Port forwarding has nothing to do with it. YOU are connecting to the server, and not vice versa, so the port must be open on the server.

Look in the logs of your instance (window events) for information about connections, connection attempts, unsuccessful authorizations, etc.
You should probably start with this. Port forwarding is clearly not needed on the router, everything should be closed on the router. But in the settings of the instance (from the Amazon side), you need to specify which port of the instance and to whom you open it. Maybe there was something wrong?
