In 2 weeks I am finishing setting up a real estate bulletin board .
Before you start promoting, you need to find the right solution: is it cheaper and easier to promote cities and regions on subdomains or in folders ?

Better subdomains. It will be easier to promote. The same Yandex will associate the domain with a specific region.
