There are 50 users, do they need CALs if they access my application that is running on the server?

If you mean Microsoft Windows Server CAL 2016, then it is used for network devices. If you have 50 computers, then you need to buy 50 licenses for them. It does not affect users in any way.
UPD: I.e. if the device was already on your network and a license was bought for it, then you don’t need to buy anything. If this is a new machine that has not been used before, then you need to buy CAL.
