Will my motherboard support ASUS TUF B450-Plus Gaming SSD M.2 Samsung 980 500GB M.2 PCIe 3.0 x4 V-NAND 3bit MLC (MZ-V8V500BW)?

I want to buy this Samsung 980 500GB SSD, but I got confused when I opened the manual for my motherboard, where it says that M.2 will work on PCI Express 3.0 x4 / SATA III. I want to make sure that M.2 will work at its declared speeds under 3000mb / s, and not at SATA speeds up to 600mb / s?

Yes, it will run at PCI Express 3.0 x4 speed. If necessary, re-read the manual again, it says that this slot supports incl. and M2 SATA SSD. But you buy NVME.
In my opinion, it is important to note that " M2 socket shares bandwidth with SATA ports

" : the bandwidth is rummaged between M2 and SATA 5/6, respectively, the latter will become unavailable.
The second M2 slot (running PCIe 2.0) seems to be shared even more.
