What is the difference between any constructors like
Readymag and Tilda and developing a site using a standard stack?
What are their main disadvantages that do not allow them to be full-fledged competition?

More junk less flexibility. This is key. Moreover, the start is fast, but when you understand what you need, you are already on the needle, and you spend resources on fighting the designer. IMHO
