First you need to understand what the connection is.

I propose then to start with a list of subsystems that you would like to check.
for example:
1. Disk subsystem.
2. Database
server 3. Application server
4. Frontend server (for example, a caching reverse proxy)
5. Quality of requests to the database (the last item on the list, but the first item to check)
