Should I use em?

All I know about em is that it is a relative unit of measure. But I'm not very sure about what, I'm guessing about the parent element, is this correct? Can you explain why em is needed? Should I use them when specifying padding and margins? And what is better to use in a responsive site: em or %?

em are anchored to the current font size.
It is convenient to use percentages to mark up an adaptive / fluid grid, and for any indents / margins when forming a block, it is often more convenient to use em or px
