Is it possible to register the port of the mail server in DNS?
There are 2 domains hosted on the same server, on the same ip.
You need to make sure that the MX record of one host leads to one port for a given ip, the MX record of another leads to another.
Is it possible to do so?

no and no reason. the mail server itself handles emails for both domains and won't get confused, don't worry.
In DNS ports? absolutely not.

By the name of the domain, sort it out and that's it.
Why invent a bicycle where it was invented a long time ago.
You can't do that.
MX records lead nowhere. MX records set the priority of the specified server in the list of candidate servers for receiving mail. Very often there is only one such record, but if there are several, then the mail will be given to the one pointed to by the MX record with the lowest priority.
Maybe it's worth reading about "virtual hosts" before you start inventing nonsense?
