Use "on-hover" mouse cursor or "hot-key": SPACE or radio button -> cheaper and easier.
In general, you can put 1 camera from 5 Mpx or more and catch the shift of the pupils from the video stream. (Better than 2 cameras at the corners of the monitor.)
Lower resolution - will not allow you to catch the change in the position of the pupils.
(you can try interpolation with zoom on a regular (built into the cover) camera 1.3, etc., but I think that this will not give the expected effect)
