I connected IPv6 to Mikrotik via Tunnelbroker, but the question, in general, is not that.

How, and where, is it correct to register a route so that certain subnets work STRICTLY via IPv4?

nowhere and in any way, the choice between ipv4 and ipv6 is made by the application. the only thing you can do is disable firewall access to certain ipv6 addresses/pools manually
