Which format is right to choose, is it necessary to optimize the image (tinypng, imageoptim and the like)?

Compression will be handled by Unity itself. All this can be done in the import twists . There it is worth paying attention to the fact that with some settings, the texture size should be a power of two (4x4, 8x4, 16x16, etc.), but not always.
It will be most convenient to work directly with the .psd sources, at the maximum resolution (as far as it is implied in the art itself).
