If you wanted to ask how to scale up a 16x16 image to 1000x1000 without antialiasing, then the solution depends on the editor you have. Let's say in IrfanView you just need to specify the Resize option instead of Resample.
In regular Microsoft Paint, you don’t need to turn anything off at all - it doesn’t know how to do anti-aliasing when you zoom in, so it simply turns one pixel into as many times as you enlarge the image.
But if you are not asking about this, but about how to enlarge a pixel art image so that the thickness of the lines on it does not increase, but only the fill area and the length of the lines increase (see the illustration below), then the answer is no way.
