How to make a subdomain in the form mysite.ru/subdomen?

Now I'm doing this on Wordpress:
There is a main mysite.ru I create
poddomen.mysite.ru on hosting and a separate database for it. Then, when the site is ready, I transfer the database and the site to another hosting/domain, etc.
I saw many people doing it like mysite.ru/poddomen
Is it more convenient or why do they do it?

mysite.ru/subdomen is not a subdomain, but a subdirectory.
poddomen.mysite.ru is a third-level domain It
's more convenient for anyone, but it's faster and more convenient to produce directories than third-level domains :)
