How to login to Joomla admin panel without error?

There is a site on Joomla 3.4 (freebsd 10, php 5.6, mysql 5.6, nginx 1.8.1) in the materials of about 70,000 articles. While in development, so caching is disabled, when I log into the admin panel for the first time in a day, the standard nginx 50x.html error always appears. I just refresh the page and after a pause ~ 20 seconds, the admin panel loads, and at first we just wait for the connection, and then the interface loads very quickly .
In the future, if you log out and log in again, the admin panel loads without errors, only after a pause. Can you tell me how to optimize this?

Look at the nginx logs immediately after the error.
