How to approach layout in a modern way?

There is a website design PSD. Didn't finish much time.
Now, right now, I want to make a specific, professional, modern layout.
What advice do you have on how to start layout and what to use?
Given the responsive layout, I'm going to do it by hand (without bootstrapa).
I think that while I haven't done it yet, technologies have appeared that make work easier.
Are there any? And yet, should I use CSS innovations, such as flex, when laying out?
