Can anyone help me get Bitcoin Core up and running?

After installing Bitcoin Core x64 (0.10.1) I couldn't run it. When I try to start it immediately throws an error.
System: Windows 8 Professional (build 9200) with the latest updates.

Well, the error clearly indicates a lack of some kind of thing. Or update .net or maybe qt needs to be installed. Although most likely the first.

MB do not have permission to write to the directory with the database? Was the old base put in, or a fresh wallet and the first launch as it is?
