Will blocking site folders via .htaccess prevent site from being hacked?

deny from all

, will this prevent the site from being hacked or not, and if not, please explain why and what else can be done?

No, it won't. This does not affect weak passwords on FTP/SSH, SLQi, file upload scripts, etc. etc.
