If the task is to recognize handwritten letters, then what recognition method / algorithm is the most relevant and gives more accurate results in this area at the moment? Is it better to use neural networks or machine learning methods without ANN? Who can advise?

In Matlab, you can use a third-party OCR library ( examples ). What's inside, in my opinion, is not important, people have probably tried different options. You just have to take a ready-made solution.
