Should an application built under .NET 4.5 but have libraries built before .NET 3.5 work on .NET 4.5?

That is, if the project has libraries from 2.0, 3.5, 4.5, then all these dotnet versions need to be installed?

Yes all. major versions are not interchangeable, minor ones are. 4.7 replaces both 4.6 and 4.5 and 4.0, 4.x does not replace 3.5 and 2.0, 3.5 does not replace 2.0.
