On the basis of what is the programming language chosen for autotesting?

I am engaged in manual testing of desktop and WEB applications. I'm looking at automation. On the basis of what is the programming language chosen for writing auto-tests? If a web project is programmed in Java, is it possible to test it using Python, for example?
