How to sign something with a user certificate?

We at work in Windows have certificates for each user.
Are there any options to open a request to the user to sign something using his certificate from a web page?
We use PHP. Is there a browser extension of some kind?

The first question is how these certificates are issued - centrally or how? If centrally, then through a Windows snap-in or third-party tools?
(The question is not idle, but related to the topic of checking the validity of a certificate)
