The connection between the host and the Mikrotik can be encrypted using IPSec. Mikrotik supports IPSec out of the box. The windows support IPSec out of the box - setup via GPO. Nixes support IPSec after some minor manipulations.
In practice, I did not try it myself - there was no need. In theory, there shouldn't be any particular problems.
And do not read what they write here in the comments, IPSec is not a VPN solution, although it is used to encrypt traffic in some of them.
The only thing - is it really necessary? If a separate vlan is considered in theory as an alternative, but for some reason it cannot be raised, then it means that encryption as such is not required. Maybe you should consider QinQ?

No way. Only VPN. Moreover, there is a vpn built into Mikrotik.

without using any of the vpn?

No way. And I strongly advise you to read about what a VPN is.
